Effects of insulin (1, 2 and 3 IU/kg, i.p.), morphine (5mg/kg, i.p.), naloxone (5mg/kg, s.c.), insulin before morphine and after naloxone were studied in the formalin test of mice. Formalin test was induced by intraplantar injection of formalin (20m, 5%). Morphine alone and insulin with morphine decreased the first phase (0-5 min) of pain. Naloxone increased the interphase (6-20 min) of niciception. Insulin and morphine decreased the second phase (21-40 min) of pain. Insulin enhanced the morphine - induced antinociception. Naloxone attenuated the suppressive effect of insulin. It seems that the antinociceptive effect of insulin in the formalin test may be dependent on the activation of opioid system.

An experiment was conducted to evaluate the impact of melatonin on scrotal circumference and semen parameters using 8 shall rams in April until June 2004 (out of season). In the April, the rams were randomly allocated in two groups. Four animal received 3 implants of melatonin tablets (totally 54 mg/ram) at the base of ear and the other 4 rams were considered as control group and kept at the identical circumstances. The scrotal circumference and semen parameters were weekly recorded over of a period of 14 weeks and the data on each variable were compared base on t-test. There was no significant differences between treatment and control group for scrotal circumference (32.8±.83 vs 32.89±0.21 cm), semen volume (0.98±0.31 vs 0.9±.12 ml/ejaculation), mass motility (3.64±0.46 vs 3.1±0.31), individual motility (70 vs 71percent), sperm concentration (2.23±0.39 vs 1.72±0.31 bilion/ml) and live sperm per cent (79 vs 82 percent ) (p>0.05). It was concluded that to use of melatonin would not improve the semen parameters in out of season reproduction assessment of shall rams.

To determine the contamination of traditional cheeses to E. coli O157:H7 this study was conducted in Chaharmahal and Bakhtiari province. Two hundred cheeses samples were obtained randomly and bacteriological examinations were done on all isolates, by culture in selective enrichment media and after that, selective plating. Suspected colonies were examined with O and H antisera for identification of serotypes of E. coli, by direct agglutination method. Two percent of unpasturized cheeses were contaminated to E. coli O157 but non of them were O157:H7. The contamination of sheep, cow and goat cheeses were 3.84%, 1.7% and 0, respectively. Seventeen percent of E. coli isolated from samples were as serotypes including, Enteropathogenic (O44, O55, O114, O125, O126, O127, O86), Enterotoxigenic (O20, O128) and Verotoxigenic (O26, O111). From the results of this study it seems that traditional cheeses could be a potent vehicle to transmit the various serotype of E. coli to human.

The present study performed to examine canola meal on morphology of thyroid gland in broiler chicks. In this experiment 300 l-day-old unsexed Ross broiler chicks assigned into 5 groups. The chicks were fed by five level of canola meal (0, 5, 10, 15 and 20%). After 42 days feeding four chicks from each pen killed and their thyroid glands removed and immediately were immersed in formaldehyde, before fixation in Bouin's solution and paraffin embedding. Each sample was then sectioned at a thickness of 7 mm, stained with haemetoxylin and eosin, and examined by light microscopy. Crimean- Congo hemorrhagic fever (CCHF) is a zoonotic arbovirus disease, which is caused by a virus of Nairovius genus and Bunyaviridae family. The present study was conducted during spring and summer 1384, and blood samples in this study were taken from dairy cow population of Khorasan provinces, including Razavi and Southern Khorasan. All together 248 blood samples belong to four cities of mentioned provinces (Mashad, Torbate heydariyeh, Sarakhs and Birjand) were studied. Prevalence of CCHF virus infection was assessed by enzyme- linked immunosorbent assay (ELISA) for immunoglobulin G (IgG) antibodies to virus. Serologic analysis of serum samples in this study showed that specific IgG against CCHF virus is present in serum samples of 9.6% of all examined samples. Among four sampled cities, anti viral IgG prevalence was highest in Sarakhs (20.7%) and Birjand had the lowest prevalence (3.3%). According to different age groups high prevalence was observed in cows over 5 years old (21.4%) and no specific antibodies found within 2- 3 years old. The presence of serological results for CCHF in dairy cattle from Khorasan province is significant and should be considered in controlling and preventive programs.

Feline leukemia virus is a retrovirus of domestic cats that occurs throughout the world. Persistent infection in the animals and persistently infected cats are at risk for developing of fatal diseases such as: lymphoma, leukemia, haematopoietic malignancies and anemia. In this study as the first time in Iran the serologic prevalence of feline leukemia virus was determined with commercial ELISA method. Sampling was performed randomly from 103 domestic and stray cats. Positive serologic result was diagnosed in five cases (4.8%). All the positive cats belonged to domestic group. There wasn't any significant differences depend on sex or age, but most Positive cases were less than 3 years old. With respect to some difficulties in vaccination protocols against feline leukemia virus, detection of the infected animals and prevention of their contact with healthy cats is considered as best method in prevention of the disease.

Staphylococcus spp., is a common bacteria in cattle mastitis. Detection of the low number of bacteria requires a sensitive method. We used multiplex polymerase chain reaction (m-PCR) assay to detect the genes encoding SEA, SEB, SEC toxins, that are specific in S. aureus bacteria. The gene of 23S rRNA that is conserved in all Staphylococcus spp., also was used. Due to the stiffness and low effective enzyme activity such as lyzozyme, proteinase k, and mutilysine on bacterial cell wall, it was lysed with liquid nitrogen, and multiplex PCR was performed after DNA extraction. The obtained results in molecular method were compared with traditional cell culture. The total number of 48 samples of non pasteurized milk was collected in Tehran province restricts from dairy cattle housing, and 4 positive samples were detected by PCR and microbial cell culture methods. The result of antibiogram showed that Ciprofloxacin, Gentamycin and Rephampine are the best choice of antibiotic for treatment of Staphylococcus spp., mastitis infectious. The accuracy of the test was monitored by using O/N cell culture serial dilution (1 to l06) of Staphylococcus spp., bacteria (OD600=0.02:107 cell). It showed that the sensitivity of PCR is 10 bacteria per ml of cells within few hours, meanwhile in bacterial blood agar cell culture; the number of 100 cells was detected after 48 hours. The result indicates that a molecular technique is more sensitive and rapid compared to traditional bacterial culture method.

On the basis of the some studies recently published on the concentration of potentially dangerous levels of mercury, a global pollutant and highly toxic element in sea fish and specially tuna fish, this work has been carried out to determine the concentration of mercury in canned tuna fish, that compose a large majority of the fish most people eat. For this reason, 30 cans, produced by different factories were randomly collected through the Tehran vicinity and the total mercury contents of all samples was measured with GBC hydride generation atomic absorption spectrometry. To better extraction of total mercury in the biologic samples, the effects of SnCl2 (reducing agant) and sample weight on the rate of mercury absorption was examined either. Statistical analysis was done applying T-test and SPSS (v.10). Based on the results, the tuna fish from different factories contained different concentrations of mercury the average of which was 146.65±63.35 ppb. Also, it was found that sample weight, and not different concentrations of SnCl2 affect absorption. Although the mercury concentrations of the samples tested was below standard or maximum residual level (MRL), due to teratogenic effects of mercury, it is strongly recommended to do screening tests on tuna fish products for domestic consumption or for export purposes.

Effects of different levels of ascorbic acid consisting of 0, 100, 200, 400, 800 and 1600 mg/kg feed were examined on some immune variables of juvenile great sturgeon (Huso huso) weighing 38 g at water temperature 20°C for 16 weeks. Significant difference were observed in level of lysozyme in fish treated 400 and 1600 mg compared with control group 8 week post-treatment (p0.05) 16 weeks post-treatment. Maximum level of leucocytes population was seen in fish treated 100 and 200 mg ascorbic acid 8 and 16 weeks post-treatment, respectively. Interaction effect of three level of male age broodstock and three type of diluents (distilled water and diluents 1 and 2) for duration of motility and eyed egg rate in rainbow trout (Oncorhynchus mykiss) have been studied. There was no significant difference in activation of sperm by control group (distilled water) in different male ages (p?0.05). Diluents 1, in 3+ year male broodstock has shown, the highest motility (98/4 s) which was significant, comparison to the other male ages. In diluents 2, 3+ year male show highest duration of motility (96/2 sec) in comparison of 2+ and +4 year male broodstock also have showed the highest motility, in comparison to 2+ and 4+ year male whereas, the 2+ year male has the lowest sperm motility. By control group, 2+ year male have shown the highest eyed egg rate (86/8 %) if compared to other male ages. Diluents 1, with 2+ year male age, showed the highest eyed egg rate which was significant, compare to other age groups. Activation of sperm by diluents 2, showed the highest eyed egg rate in 2+ and 3+ year male age broodstock.
