Samples: Eighteen Pasteurella multocida isolates obtained from poultry during 10 years (1988-1998) from Mazandaran province. Procedure: Twenty five to thirty /lg of bacterial protein was electrophoresed in a discontinuous polyacrylamide gel electrophoresis system. The concentration of acrylamide in resolving and stacking gels were 12% and 4.5% respectively. Minimum lethal dose of each protein type was determined by IP injection of several bacterial dilutions to groups (n= 3) of Balb/c mice. To study the cross- protection, four groups of 10 mice were immunized with vaccinal strain. The immunized mice together with control groups were challenged with homologous and heterologous protein type isolates. Results: The electrophoretic pattern of the isolates contained over 30 polypeptide bands ranging from 24 to 174 kda. The overall . protein pattern of the isolates were similar. The main difference was in the position of a major protein band, which is known as protein H. The position of this band was presented in 38,36.5 and 34 kda. Based on this difference the isolates were classified as protein types I, II and III respectively. Immunization of mice with the vaccinal strain (protein type I) protected the animals against homologous challenge. Conclusion: According to results of this study, preparation of a polyvalent vaccine containing different protein types are suggested to prevent fowl cholera in Mazandaran province. J. Fac. Vet. Med. Tehran. 58,3: 197-201, 2003.}, keywords = {Cross-immunity,Pasteurella multocida,Protein fingerprinting,virulence}, title_fa = {مطالعه الگوی پروتئینی پاستورلامولتوسیدا جدا شده از طیور و مقایسه حدث پروتئین تیپ های شناسایی شده در موش}, abstract_fa = {هدف : تعیین ویژگی الگوی پروتئینی و طبقه‌بندی نمونه‌های پاستورلا مولتوسیدا جدا شده از طیور استان مازندران، بررسی ایمنیت متقاطع و نیز مقایسه حدت پروتئین تیپهای شناسایی شده در مدل موشی. نمونه‌ها: تعداد 18 نمونه پاستورلا مولتوسیدا جدا شده از طیور استان مازندران که در طی ده سال (1388-1368) در موسسه رازی جدا شده‌اند. روش : مقدار 30-25 میکروگرم پروتئین خام تهیه شده از جدایه‌ها در سیستم ناپیوسته در ژل سدیم دو دسیل سولفات پلی آکریل آمید الکتروفورز گردید. غلظت آکریل آمید در ژل جدا کننده 5/12 درصد و در ژل متراکم کننده 5/4 درصد بود. حداقل دوز کشنده هر تیپ پروتئینی با تزریق رقتهای مختلف باکتری پاستورلا مولتوسیدا به گروههای سه تایی موش از طریق داخل صفاقی تعیین گردید. چهار گروه ده‌تایی موش ایمن شده با سویه واکسینال به همراه چهار گروه ده‌تایی به عنوان کنترل برای بررسی ایمنی متقاطع با سایر تیپهای پروتئینی مورد استفاده قرار گرفت. نتایج : بیش از 30 باند پلی پپتیدی در فاصله بین 24 تا 174 کیلو دالتون مشاهده شد. شکل کلی الگوهای پروتئینی جدایه‌ها بسیار به هم شباهت داشت. اختلاف عمده بین آنها در محل قرار گرفتن یکی از باندهای اصلی موسوم به پروتئین H بود. این باند در جدایه‌های مختلف در سه موقعیت 38، 5/36 و 34 کیلو دالتون قرار داشت که بر همین اساس جدایه‌ها را در سه پروتئین تیپ (III , II , I) جای داد. ایمن‌سازی گروه‌های موش با سویه واکسینال (پروتئین تیپ I) توانست آنها را در مقابل چالش با پروتئین تیپ همنوع (همولوگ) حفاظت نماید. نتیجه‌گیری : بر اساس یافته‌های این مطالعه تهیه یک واکسن پلی والان متشکل از هر سه پروتئین تیپ جهت پیشگیری از وبای مرغان در این منطقه پیشنهاد می‌گردد.}, keywords_fa = {الگوی پروتئینی,ایمنیت متقاطع,پاستورلامولتوسیدا,حدث}, url = {https://jvr.ut.ac.ir/article_11904.html}, eprint = {https://jvr.ut.ac.ir/article_11904_84b9feaed1813cba271a52e42794642e.pdf} }
