The use of flow cytometrictechniques and monoclonal antibodies are appropriate methods in the diagnosis ofhematopoietic malignancies. OBJECTIVES: The purpose of the present study is to determinecell-surface antigens for various developmental stages of feline bone marrow cells inhematopoietic disorders using flow cytometric. METHODS: In this study, bone marrow cellsfrom 4 cats with hematopoietic disorders and 2 clinically healthy cats, were labeled with 5types of anti-feline MAbs included: CD21-like (Cr-Br), T lymphocyte subpopulation, CD-172a, Granulocyte, Pan-Leukocyte (CD45-like) and then analyzed using flow cytometric.RESULTS: The results revealed changes in immunophenotyping and light scatter propertiescompared with normal cases. The percentage of CD45, Granulocyte and CD172a markers inthe bone marrow of a cat with erythroleukemia were lower compared with normal bonemarrow. In a cat with myelodysplastic syndrome, scatter plot indicated an increase in theimmature myeloid cells and a decrease in mature myeloid cells. CONCLUSIONS: It wasconcluded that cytological examination of bone marrow with studying dispersion studies oncells using flow cytometric and usage of a panel of antibodies such as CD21-like(Cr-Br), Tlymphocyte subpopulation, CD-172a, Granulocyte, Pan-Leukocyte (CD45-like) couldsupport the diagnosis of feline hematopoietic abnormalities. }, keywords = {Cat,flow cytometric,hematopoietic disorders,immunophenotyping}, title_fa = {بررسی ایمنوفنوتایپینگ و فلوسایتومتری سلول‌های مغز استخوان در گربه‌های مبتلا به اختلالات هماتوپویتیک}, abstract_fa = {زمینه مطالعه: بررسی سیتولوژیک مغز استخوان گربه به دلیل تعداد زیاد سلول‌ها و مراحل مختلف رشد تا حدودی پیچیده است. تکنیک‌های فلـوسـایتومتری و آنتی‌بادی‌های منوکلونال روش مناسبی به ویژه در تشخیص بدخیمی‌های هماتوپویتیک می‌باشند. هدف: بررسی سلولی مغز استخـوان گـربه‌های مبتلا به اختلالات هماتوپویتیک با استفاده از آنتی بادی‌های منوکلونال با روش فلوسایتومتری. روش کار: در این مطالعه سلول‌های مغز استخوان‌ 4 گربه با اختلالات هماتوپویتیک و دو گربه سالم از نظر کلینیکی با روش فلوسایتومتری مقایسه شدند.  آنتی‌بادی‌هایی مورد استفاده در این بررسی شامل -like (Cr-Br)21CD‌، ‌T lymphocyte subpopulation‌،   ‌a172CD-‌، گرانولوسیتی و پن‌لکوسیتی -like)45(CD‌ بودند. نتایج: فلوسایتومتری مغز استخوان گربه‌های مبتلا نشان دادند که تغییراتی  در خصوصیات ایمنوفنوتایپی و پراکنش نوری (Light Scatter) در مقایسه با موارد کنترل وجود داشتند. در گربه مبتلا به اریترولوکمی بیان مارکر ‌45CD‌، گرانولوسیتی و ‌a172CD‌ کمتر از موارد کنترل (نرمال) بودند. در گربه مبتلا به میلودیس‌پلازی، در بررسی نمودار نقطه‌ای، افزایش سلول‌های نابالغ میلوییدی و کاهش سلول‌های بالغ میلوییدی مشاهده شد. نتیجه گیری‌نهایی: این نتایج بیانگر آن است که همراه با بررسی سیتولوژی سلول‌های مغز استخوان، مطالعه خصوصیات پراکنشی سلــول‌هــا در فلــوســایـتــومـتــری وهـمـچـنـیــن اسـتـفــاده ازآنـتــی بــادی‌هــای  ‌-like21CD‌، گــرانـولـوسیـت،   پـن لکـوسیتـی ‌45CD‌،                                                      T lymphocyte subpopulation و a172CD‌‌ می‌توانند در تشخیص اختلالات هماتوپویتیک گربه تکمیل کننده باشد.  }, keywords_fa = {گربه,فلوسایتومتری,‌ اختلالات هماتوپویتیک,‌ایمنوفنوتایپینگ}, url = {https://jvr.ut.ac.ir/article_36711.html}, eprint = {https://jvr.ut.ac.ir/article_36711_942cc25cf56134d2484d90842c542817.pdf} }
