OBJECTIVES: The purpose of present study was detection of gyrA gene (quinolone resistance) in the Y. ruckeri bacterium. METHODS: In this study fish were evaluated in average size 8-12 cm from six rainbow trout farms in Chahar Mahal va Bakhtiyari province (Iran). In each farm 10 fish (totally 60) suspected to yersinioisis were randomly selected; sampling was done from lower part of intestine and cultured on Trpticase Soy Agar (TSA). The mediums were transferred to incubator and kept at 22 °C for 48 hours. Pure colonies which are grown on the mediums were tested by catalase, oxidase and gram staining, then those of gram-negative, catalase positive and  oxidase negative were diagnosed, and cultured on Waltman- Shots medium (as specific medium for Y. ruckeri). These mediums were incubated at 22 °C for 48 h. Colonies that were grown were tested by PCR method for Y.ruckeri detection. Then, in the identified strains of Y.ruckeri gyrA gene were detected by PCR test. RESULTS: The results of bacteriological, biochemical and molecular tests showed that three cases out of total isolates were identified as Y. ruckeri. In all isolates of Y. ruckeri, gyrA gene was identified by molecular test. CONCLUSIONS: Identification of quinolone resistance gene in Y. ruckeri isolates can be the reason of low efficacy of these classes of antibiotics in the aquaculture. ِTherefore, the policy of treatment should be changed specially in enteric red mouth disease.}, keywords = {bacterial resistance,gyrA,PCR,Rainbow trout,Yersinia ruckeri}, title_fa = {ردیابی مولکولی ژن مقاوم به کینولونها (gyrA) درجدایههای باکتری یرسینیاراکری با آزمون PCR}, abstract_fa = {زمینه مطالعه: یرسینیاراکری عامل بیماری دهان قرمزآنتروباکتریایی یا یرسینیوز (یکی از بیماریهای مهم آزاد ماهیان پرورشی) است. هدف: مطالعه حاضر باهدف ردیابی ژن مقاوم به کینولون‌ها (gyrA) در باکتری یرسینیاراکری صورت پذیرفت. روش کار: به منظور انجام این تحقیق شش مزرعه پرورش ماهی قزل‌آلای رنگین‌کمان استان چهار محال و بختیاری مورد بررسی قرار گرفت. به طوریکه از هر مزرعه 10 عدد و در مجموع 60 عدد ماهی مشکوک به بیماری با اندازه cm‌12-8 به طور تصادفی انتخاب و اقدام به نمونه برداری از انتهای روده آنها و کشت بر روی محیط TSAر(Tripticase Soy Agar) گردید. سپس محیط‌ها به انکوباتور انتقال و بعد از 24 ساعت گرم‌خانه گذاری در دمای C° 22، برروی پرگنه‌های رشد یافته آزمون‌های کاتالاز، اکسیداز و رنگ گرم انجام شده وآنهایی که گرم منفی، کاتالاز مثبت و اکسیداز منفی بودند به محیط کشت اختصاصی واتمن شاتس منتقل شدند. پس از گرم‌خانه گذاری در دمای C°22 به مدت 48 ساعت از پرگنه‌های رشد یافته تست PCR انجام شد. در مرحله بعد بر روی باکتری‌های یرسینیا راکری شناسایی شده ردیابی ژن gyrA با استفاده از آزمون PCR صورت پذیرفت. نتایج: نتایج آزمون‌های باکتری شناسی، بیوشیمیایی و مولکولی تحقیق حاضرنشان داد که 3 نمونه از کل باکتری‌های مورد بررسی، باکتری یرسینیا راکری بوده و در همه آنها ژن gyrA ردیابی گردید. نتیجه‌گیری‌نهایی: شناسایی ژن مقاومت به کینولون‌ها در باکتری یرسینیا راکری می‌تواند دلیلی بر کاهش کارایی درمانی این دسته آنتی بیوتیک‌ها در مزارع پرورش ماهی باشد و لذا بایستی سیاست کنترل و درمان بیماریهای عفونی به ویژه دهان قرمزآنتروباکتریایی تغییر کند.}, keywords_fa = {gyrA,PCR,یرسینیا راکری,قزل آلای رنگین کمان,مقاومت باکتریایی}, url = {https://jvr.ut.ac.ir/article_57400.html}, eprint = {https://jvr.ut.ac.ir/article_57400_6fdfe0267f24ad221bca736c1423840d.pdf} }
