The mechanisms involved in AIA are not fully understood but it seems that inhibition of cyclooxygenase (COX) can play a role in such attacks.               Objectives: This study aimed to compare the effects of aspirin-like drugs on airway responsiveness in an animal model of allergic asthma. Methods: 42 male guinea pigs (250-300 g) were randomly divided into seven groups: control, allergic, ketoprofen, indomethacin, celecoxib, and aspirin (high and low doses). All animals except control group were sensitized by i.p. injections of ovalbumin and Al (OH)3 suspensions and then challenged by inhalation of ovalbumin solution on day 18. The animals received three oral doses of the aforementioned drugs or normal saline in control and allergic groups with 12 hr intervals within 24 hr before ovalbumin challenge. On day 19, animals were euthanized and the responsiveness of isolated tracheas was studied using cumulative doses of histamine and acetyl choline in an organ bath. Results: Indomethacin and ketoprofen significantly increased the trachea responsiveness to acetyl choline and histamine at all concentrations. Emax values for acetyl choline and histamine in indomethacin were respectively (3.5±0.1, 3.5±0.1 g) and ketoprofen (5.1±0.2, 4.3±0.1) groups were greater than those of control (1.5±0.1, 1.5±0.1), allergic, celecoxib, high dose aspirin and low dose aspirin groups (p<0.05). Conclusions: It was concluded that indomethacin and ketoprofen (but not aspirin and celecoxib) can exacerbate the asthmatic conditions in this animal model.}, keywords = {airway responsiveness,allergy,aspirin-induced asthma,cyclooxygenase inhibitor drugs}, title_fa = {مقایسه اثرات مهارکنندههای منتخب آنزیم سیکلواکسیژناز بر پاسخدهی راههای هوایی به هیستامین و استیل کولین در خوکچههای هندی مبتلا به آلرژی تجربی}, abstract_fa = {زمینه مطالعه: آسم ناشی از آسپرین (AIA) با علائمی همچون انقباض حاد برونش‌ها در حدود 10% از افراد مبتلا به آسم بدنبال مصرف آسپرین یا داروهای مشابه بروز می‌کند. مکانیسم‌های ایجاد AIA بطور کامل شناخته نشده اند، اما به نظر می‌رسد مهار آنزیم سیکلواکسیژناز در بروز آن دخیل باشد. هدف: مقایسه اثرات داروهای شبه آسپیرین بر پاسخ‌دهی مجاری هوایی در خوکچه هندی مبتلا به آسم آلرژیک تجربی. روش کار: 42 عدد خوکچه هندی نر (300g-250) بطور تصادفی در هفت گروه تقسیم شدند: گروه کنترل، آلرژیک، کتوپروفن، ایندومتاسین، سلکوکسیب و آسپیرین (با دوز بالا و پایین). در همه حیوانات بجز گروه کنترل، بوسیله تزریق داخل صفاقی اوآلبومین و هیدروکسید آلومینیوم آلرژی ایجاد شد و چالش با اوآلبومین استنشاقی در روز 18 صورت گرفت. در طی 24 ساعت قبل از چالش، حیوانات سه دوز خوراکی از داروهای فوق و یا محلول کربس هنسلیت در گروه‌های کنترل و آلرژیک، هر 12 ساعت یکبار دریافت کردند (دوز آخر درست نیم ساعت قبل از چالش تجویز گردید). در روز 19 با ایجاد مرگ آرام در حیوانات، پاسخ‌دهی قطعات مجزای نای آنها با استفاده از دوزهای تجمعی هیستامین و استیل کولین در حمام بافت مطالعه گردید. نتایج: ایندومتاسین و کتوپروفن بطور معنی‌دار پاسخ‌دهی نای را در تمامی غلظت‌های هیستامین و استیل کولین افزایش دادند. میزان Emax برای استیل کولین و هیستامین برحسب گرم به ترتیب در گروه‌های ایندومتاسین (1/0±5/3 و 1/0±5/3) و کتوپروفن (1/0± 3/4 و 2/0±1/5) بیشتر از گروه‌های کنترل (1/0± 5/1 و 1/0±5/1)، آلرژیک، سلکوکسیب، آسپرین با دوز بالا و آسپیرین با دوز پایین بود  (0.05>p).  نتیجه‌گیری نهایی: این مطالعه نشان داد که ایندومتاسین و کتوپروفن (و نه آسپرین و سلکوکسیب) میتوانند موجب افزایش پاسخ‌دهی مجاری هوایی و تشدید علایم آسم در این مدل حیوانی شوند.}, keywords_fa = {پاسخدهی مجاری هوایی,آلرژی,آسم ناشی از آسپیرین,داروهای مهارکننده سیکلواکسیژناز}, url = {https://jvr.ut.ac.ir/article_57919.html}, eprint = {https://jvr.ut.ac.ir/article_57919_2cc9f5a59660629a30633e889e9a10ca.pdf} }
