OBJECTIVES: The aim of this study was to evaluate the prevalence of the genes encoding collagen binding (cna) and fibronectin binding (fnb) adhesion factors in Staphylococcus aureus isolates from food and clinical specimens. METHODS: During the time period 2010-2013, a total of 38 isolates of Staphylococcus aureus from clinical specimens and 32 isolates from food samples were collected. All isolates were identified biochemically and subjected to DNA extraction. The accuracy of DNA extraction from each isolate was confirmed by PCR amplification of aroA gene and then the presence of cna and fnb genes in the extracted DNAs was assessed by PCR, using the specific primers. RESULTS: The results showed that among 38 isolates from clinical specimens, 15 (39.5%) & 32 (84.2%) and among 32 isolates of food origin 10 (31/2%) and 16 (50%) isolates had cna and fnb genes respectively. Thirteen clinical (34.21%) and 6 food isolates (18.7%) were positive for both fnb and cna genes and 4 clinical (10.5%) and 12 food isolates (37.5%) were lacking both genes. CONCLUSIONS: According to the results, it is concluded that, although these adhesion genes are not present in all Staphylococcus aureus clinical isolates, but their prevalence is high and using the products of these genes in vaccines may be effective in protecting against infections caused by this bacterium.  }, keywords = {adhesion genes,cna and fnb,Staphylococcus aureus}, title_fa = {مقایسه شیوع ژنهای کد کننده پروتئینهای متصل شونده به کلاژن (cna) و فیبرونکتین (fnb) در جدایههای استافیلوکوکوس آرئوس بدست آمده از مواد غذایی و موارد درمانگاهی}, abstract_fa = {زمینه مطالعه: استافیلوکوکوس آرئوس (S. aureus) ژن‌های حدت متنوعی دارد که در بروز عفونت‌های ناشی از این عامل نقش دارند. هدف: هدف از این تحقیق، بررسی فراوانی ژن‌های مربوط به عوامل چسبان پروتئین متصل شونده به کلاژن (cna) و پروتئین متصل شونده به فیبرونکتین (fnb) در جدایه‌های استافیلوکوکوس ارئوس‌ حاصل از مواد غذایی و عفونت‌های درمانگاهی بود. روش‌کار: در محدوده زمانی سال‌های 1392-1389، تعداد 38 جدایه استافیلوکوکوس آرئوس از نمونه‌های درمانگاهی و 32 جدایه نیز از نمونه‌های مواد غذایی جداسازی و جمع‌آوری گردید. تمام جدایه‌ها با روش‌های بیوشیمیایی شناسایی‌شده و مورد استخراج DNA قرار گرفتند. صحت استخراج DNA با تکثیر ژن aroA به‌وسیله PCR در هر یک از جدایه‌ها مورد تأیید قرار گرفت و سپس حضور ژن‌های cna و fnb در DNA هر یک از جدایه‌ها با PCR به کمک پرایمرهای اختصاصی این ژن‌ها ردیابی شد. نتایج: نتایج نشان داد از میان 38 جدایه به‌دست‌آمده از نمونه‌های درمانگاهی تعداد 15 (5/39%) و 32 (2/84%) جدایه و از میان 32 جدایه با منشأ مواد غذایی تعداد 10 (2/31%) و 16 (50%) جدایه به‌ترتیب واجد ژن‌های cna و fnb بودند. تعداد 13 جدایه‌ درمانگاهی (21/34%) و 6 جدایه با منشأ مواد غذایی (7/18%)، واجد هر دو ژن چسبان cna و fnb بودند و تعداد 4 جدایه درمانگاهی (5/10%) و 12 جدایه با منشأ مواد غذایی (5/37%) فاقد هر دو ژن بودند. نتیجه‌گیری نهایی: با توجه به نتایج حاصل از این مطالعه می‌توان گفت، اگرچه این عوامل چسبان در همه جدایه‌های درمانگاهی استافیلوکوکوس اورئوس بیان نمی‌شوند، اما شیوع آن‌ها بالا بوده و ممکن است، استفاده از فراورده این دو ژن در واکسن‌های ضد این عامل در جلوگیری از عفونت‌های حاصل از این باکتری مؤثر واقع شود.  }, keywords_fa = {عوامل حدت,fnb,cna,استافیلوکوکوس آرئوس}, url = {https://jvr.ut.ac.ir/article_58732.html}, eprint = {https://jvr.ut.ac.ir/article_58732_709b47a69433dcfab355bb8bf9b85c5e.pdf} }
