Phospholipase D (PLD) is a potent exotoxin produced by C. pseudotuberculosis and it has been considered as the major virulence factor for this bacterium, possibly contributing to the spread of the bacteria from the initial site of infection to secondary sites within the host. Heat shock proteins (HSPs) are important candidates for the development of vaccines because they are usually able to promote both humoral and cellular immune responses in mammals. OBJECTIVES: The aim of this study was the cloning and expression of the PLD and HSP60 genes of C. pseudotuberculosis, used subsequently to evaluate the protectivity of these recombinant proteins for vaccine development against this bacterium. METHODS: PLD and HSP60 genes were cloned into pMAL-c2X vector and recombinant plasmids construct was transformed to DH5 strain of E. coli. Expression of the proteins was shown by SDS-PAGE and accuracy of the cloned genes was confirmed by nucleotide sequence analysis. RESULTS: The transformed E. coli strain DH5 expressed PLD and HSP60 proteins effectively. The expressed fusion protein was found almost entirely in the soluble form. CONCLUSIONS: In the following studies the immunogenicity and protectivity of these recombinant proteins against C. pseudotuberculosis infections can be assessed.}, keywords = {Corynebacterium pseudotuberculosis,phospholipase D,pheat-shock protein,Cloning}, title_fa = {تولید پروتئینهای نوترکیب فسفولیپاز D و پروتئین شوک حرارتی کورینه باکتریوم سودوتوبرکلوزیس}, abstract_fa = {زمینه مطالعه: بیماری لنفادنیت پنیری ناشی از کورینه‌باکتریوم سودوتوبرکلوزیس یکی از مهم‌ترین بیماری‌های گوسفند و بز است که منجر به ضرر‌های اقتصادی دام‌پروران می‌شود. فسفولیپاز Dا(PLD) این باکتری یک اگزوتوکسین قوی است که به‌عنوان یک عامل حدت اصلی آن در نظر گرفته می‌شود و احتمالاًً در انتشار باکتری از محل عفونت به سایر قسمت‌های بدن میزبان مشارکت دارد. پروتئین‌های شوک حرارتی (HSPs) کاندیدا‌های خوبی برای تهیه واکسن‌ها هستند چون هم ایمنی هومورال و هم ایمنی سلولی را تحریک ‌می‌کنند. هدف: مطالعه حاضر باهدف تولید پروتئین‌های نوترکیب PLD و HSP60 کورینه‌باکتریوم سودوتوبرکلوزیس جهت فراهم شدن زمینه‌ی بررسی توان محافظت‌کنندگی و ساخت واکسن ضد این باکتری صورت گرفت. روش کار: ژن‌های PLD و HSP60، در پلاسمید pMAL-c2X کلون گردیدند و در اشرشیا‌کلی سویه DH5 ترانسفورمه شدند. بیان پروتئین‌ها با استفاده از روش SDS-PAGE بررسی‌شده و صحت ردیف نوکلئوتیدی محصولات کلون شده با تعیین توالی نوکلئوتیدی مورد تأیید قرار گرفتند. نتایج: نتایج نشان داد که اشرشیا‌کلی سویه DH5 ترانسفورمه شده به‌خوبی پروتئین‌های فوق را بیان می‌نماید. پروتئین‌های نوترکیب بیان‌شده تقریباً به‌طور کامل محلول بودند. نتیجه‌گیری نهایی: با توجه به امکان تولید پروتئین‌های نوترکیب فوق، در مطالعات بعدی می‌توان ایمنی‌زایی و محافظت‌کنندگی آن‌ها در برابر عفونت‌های ناشی از کورینه باکتریوم سودوتوبرکلوزیس را موردسنجش قرارداد.}, keywords_fa = {کورینه باکتریوم سودوتوبرکلوزیس,فسفولیپاز D,پروتئین شوک حرارتی (HSP60),کلونینگ}, url = {https://jvr.ut.ac.ir/article_61367.html}, eprint = {https://jvr.ut.ac.ir/article_61367_4a004a8b5fe24718ccd8a4ffbaefee12.pdf} }
