The mucus layer on the surface of the fish body contains anti-microbial combination that provides the first layer of defense against pathogens. The mucus is released by some of the epidermis cells which are called goblet cells and it mostly contains the mucin and other glycoproteins. OBJECTIVES: Histomorphometrical, Histochemical and Electron Microscopic Studies of Goblet Mucous Cells in Different Regions of Argyrosomus Hololepidotus Epidermis. METHODS: In this study, six Argyrosomus hololepidotus are used and the structure of the fish’s skin was studied. For doing this microscopic study, the sampling was done on dorsal Regions of fish with a thickness of 0.5µ then they were stained with H & E, PAS, AB (PH = 2.5) and AB (PH = 2.5)-PAS. For electron microscopic study, the samples after primary and post-fixation were dehydrated and were embedded in resin. Then, thin sections 50 μm were prepared and stained with uranyl acetate. RESULTS: Argyrosomus hololepidotus fish has maximum goblet cells in ventral and dorsal skin and minimum numbers of goblet cells were seen in tail skin in 100 µm length of epidermis. There were goblet mucous cells containing mucous in the Argyrosomus Hololepidotus epidermal that thier numbers were different in difference areas but mucus components were similar in different areas and they reacted positively to PAS and AB dyes with PH = 2.5.The electron microscopic results of this study were showed that goblet cells immigrate in thickness of epidermis and they include mucosal drops. CONCLUSIONS: There are goblet mucus cells in all parts of Argyrosomus Hololepidotus Epidermis and they have similar mucus nature.}, keywords = {Histochemistry,Skin,Argyrosomus hololepidotus,Epidermis,Goblet Mucous Cell}, title_fa = {مطالعه هیستومرفومتریک، هیستوشیمی و الکترون میکروسکوپی سلول های جامی نواحی مختلف اپیدرم میش ماهی Argyrosomus hololepidotus}, abstract_fa = {زمینه مطالعه: پوست اولین سد دفاعی در مقابل محیط خارجی بوده و اعمال فیزیولوژیکی طبیعی داخل بدن را امکان پذیر می‌‌کند. لایه موکوس روی سطح بدن ماهی حاوی ترکیبات ضد میکروبی است که اولین لایه دفاعی بر علیه پاتوژن‌ها را فراهم می‌کند. موکوس به وسیله برخی از سلول‌های اپیدرم به نام سلول‌های جامی‌شکل ترشح می‌شود و عمدتا شامل موسین و دیگر گلیکوپروتئین‌ها می‌باشد. هدف: مطالعه هیستومرفومتریک، هیستوشیمی و الکترون میکروسکوپی سلول های جامی نواحی مختلف اپیدرم میش ماهی. روش کار: در این پژوهش تعداد 6 عدد میش ماهی مورد استفاده قرار گرفت و برای انجام مطالعات میکروسکوپی از ناحیه پشتی برش‌هایی به ضخامت µ 5/0 تهیه و مورد رنگ آمیزیH&E ، PAS ، AB(PH=2.5) و AB(PH=2.5)-PAS قرار گرفتند. برای مطالعات میکروسکوپ الکترونی پس از ثبوت اولیه و ثانویه و آبگیری در داخل رزین آغشته گردیده و پس از تهیه برش‌های فوق نازک nm50 در داخل یورانیل استات رنگ آمیزی گردیدند. نتایج: میش ماهی در نواحی شکمی و پشتی دارای بیشترین و در نواحی دم دارای کمترین تعداد سلول موکوسی جامی شکل در mµ100 از طول اپیدرم است. سلول‌های موکوسی جامی شکل در تمام نواحی وجود داشته و تعداد آنها در نواحی مختلف متفاوت است اما ماهیت موکوسی این سلول‌ها در نواحی مختلف یکسان است و به رنگ‌های PAS و AB با PH=2.5 واکنش مثبت نشان می دهند. نتایج الکترون میکروسکوپی نشان داد که سلول‌های جامی ترشح کننده موکوس در ضخامت اپیدرم مهاجرت می‌کنند و حاوی قطرات موکوس می‌باشند. نتیجه گیری نهایی: سلول‌های موکوسی جامی شکل در اپیدرم تمام نواحی میش ماهی جود داشته و دارای ماهیت موکوسی یکسانی است.}, keywords_fa = {هیستوشیمی,پوست,میش ماهی,اپیدرم,سلول موکوسی جامی شکل}, url = {https://jvr.ut.ac.ir/article_68392.html}, eprint = {https://jvr.ut.ac.ir/article_68392_69f7fedcd99fb6327f3dd21da9952bfb.pdf} }
