OBJECTIVES: This study was investigated for single nucleotide polymorphisms of MHC region related to the immune system in commercial broiler and layer chickens. METHODS: One hundred blood samples were taken from commercial broiler and layer chickens and genomic DNA was extracted by salting out method. The allelic polymorphisms were investigated in B-L, B-F and B-G loci using PCR-RFLP and MspI enzyme. RESULTS: For two commercial broiler and laying populations, in the 374 bp locus of B-L, only BB genotype was detected but in the 1048 bp locus of B-F, two genotypes of CG and GG were identified in broiler chickens. The C allele contained four bands of 515, 410, 75 and 47 bp, and the G allele with five bands of 410, 302, 213, 75 and 47 bp. In B-G (401 bp) locus, three genotypes of MM, MN and NN and two alleles of M with one band (401 bp) and N with two bands (350 and 51 bp) were identified. In total populations, the Shannon information index was calculated to be 0.45 and 0.73 in markers loci of B-F and B-G, and the fixation index values were -0.20 and 0.34, respectively. The highest observed heterozygosity index for B-F and B-G loci was 0.34 and 0.23, respectively. CONCLUSIONS: Considering the confirmation of the presence of polymorphism in two loci of the B-F (in commercial broiler population) and B-G (in commercial broiler and layer populations), these sites can be used as genetic marker in breeding programs to increase resistance to diseases.}, keywords = {Commercial chicken,Immune system,MHC,PCR-RFLP,Polymorphism}, title_fa = {مطالعه چند شکلی های تک نوکلئوتیدی ناحیه مجتمع عمده پذیرش بافتی مرتبط به سیستم ایمنی در مرغ های تجاری گوشتی و تخم گذار}, abstract_fa = {زمینۀ مطالعه: ناحیه  مجتمع عمده پذیرش بافتی (MHC) مرغ‌ها در صفات تولیدی، پاسخ‌های ایمنی، مقاومت به بیماری‌های عفونی و روابط فیلوژنتیکی، دارای اهمیت زیادی است. هدف: این تحقیق به منظور بررسی چند­شکلی­های تک نوکلئوتیدی ناحیه ژنی MHC مرتبط به سیستم ایمنی در مرغ­های تجاری گوشتی و تخم­گذار انجام گرفت. روش‌کار: در این تحقیق یک‌صد نمونه خون از مرغ­های تجاری گوشتی و تخم­گذار اخذ و DNA­ی ژنومی به روش شستشوی نمکی استخراج شد. چند­شکلی­های­ آللی در جایگاه‌های ژنی B-L، B-F و B-Gبا استفاده از تکنیک PCR-RFLP و آنزیم­ Msp I بررسی شد. نتایج: برای دو جمعیت تجاری گوشتی و تخم­گذار برای جایگاه تکثیری 374 جفت بازی B-L، تنها ژنوتیپ BB اما در جایگاه ژنی 1048 جفت بازی B-F، دو ژنوتیپ CG و GG فقط در مرغ­های تجاری گوشتی شناسایی شد. در این جایگاه ژنی آلل C شامل باندهای 515، 410، 75 و 47 جفت بازی و آلل G دارای باندهای 410، 302، 213، 75 و 47 جفت بازی بودند. در جایگاه ژنی 401 جفت بازی B-G، سه ژنوتیپ MM، MN و NN و دو آلل M شامل یک باند 401 جفت بازی و آلل N دارای دو باند 350 و 51 جفت بازی مورد شناسایی قرار گرفت. در کل جمعیت­ها، شاخص اطلاعات شانون در دو جایگاه ­ژنی B-F و B-G به ترتیب 45/0 و 73/0 و شاخص تثبیت به ترتیب 20/0- و 34/0 محاسبه شد. بیشترین مقدار شاخص هتروزیگوسیتی مشاهده ‌شده برای جایگاه­های ژنی B-F و B-G به ترتیب 34/0 و 23/0 برآورد شد. نتیجه‌گیری نهایی:با توجه به تأیید وجود چندشکلی در دو جایگاه ژنی B-F(در جمعیت مرغ تجاری گوشتی) و B-G(در جمعیت­های مرغ­های تجاری گوشتی و تخم­گذار)، می‌توان از این جایگاه‌ها به ‌عنوان نشانگر ژنتیکی در برنامه­های اصلاح نژادی جهت افزایش مقاومت به بیماری­ها‌ استفاده کرد.}, keywords_fa = {چندشکلی,سیستم ایمنی,مجتمع عمده‌ پذیرش بافتی,مرغ تجاری,PCR-RFLP}, url = {https://jvr.ut.ac.ir/article_74137.html}, eprint = {https://jvr.ut.ac.ir/article_74137_0ba5361d124cc9c0bee2e783b8a93ebe.pdf} }
