Due to certain factors, such as limited connective tissue available, potentiated growth of excessive granulation tissue, risk of contamination, and poor response to common treatments, healing of these wounds becomes a major problem for veterinarians on a number of occasions. Application of Mesenchymal Stem Cells (MSC) for enhancing wound healing has received a great deal of scientific attention. Among the MSCs, those derived from adipose tissue are frequently used owing to their availability, large number of cells after the primary harvest, and the capacity to differentiate to different cell lines.OBJECTIVES: The current study aimed to evaluate type 1 and 3 collagen genes expression in horse distal limb wounds treated via adipose-derived Mesenchymal Stem Cells and its comparison with bone marrow-derived stem cells.METHODS: After treatment of the experimental open wounds created in the distal limbs of four horses via autologous MSCs, real-time PCR was used for evaluating and comparing the expression of type I and III collagen genes in the healing wounds.RESULTS: Significant differences in the expression of type I and III collagen genes were observed between the treatment groups. Despite the fact that the greatest collagen genes expression belonged to bone marrow-derived MSCs, no significant differences were seen with adipose-derived MSCs.CONCLUSIONS: Owing to the advantages and an acceptable performance, adipose-derived MSCs could be considered as a novel approach to enhancing limb wound healing in horses.}, keywords = {Wound healing,equine,Mesenchymal stem cells,bone marrow,adipose tissue}, title_fa = {ارزیابی بیان ژن‌های مربوط به کلاژن‌های تیپ 1 و 3 در زخم باز التیام یافته اندام حرکتی اسب با استفاده از سلول‌های بنیادی خودی مشتق از بافت چربی و مقایسه آن با سلول‌های بنیادی مشتق از مغز استخوان}, abstract_fa = {زمینۀ مطالعه: زخم‌های باز اندام حرکتی یکی از شایع‌ترین مشکلات در اسب‌ها می‌باشد که به دلایلی نظیر کمبود بافت نرم، رشد سریع بافت گرانوله اضافه، آلودگی زیاد و پاسخ ضعیف به درمان‌های رایج، ترمیم آن‌ها معضل بزرگی برای دامپزشکان است. استفاده از سلول‌های بنیادی مزانشیمی جهت ترمیم زخم‌ها از روش‌هایی است که به تازگی توجه محققین را به خود جلب کرده است. در این میان سلول‌های بنیادی مشتق از بافت چربی به عللی نظیر سهولت تهیه، تعداد بسیار زیاد سلول‌های بنیادی حاصله در استحصال اولیه و قابلیت تمایز به رده‌های مختلف سلولی کاربرد فراوان یافته‌اند.هدف: مطالعه حاضر به منظور ارزیابی بیان ژن‌های کلاژن‌های تیپ 1 و 3 در روند التیام زخم باز اندام حرکتی اسب با استفاده از سلول‌های بنیادی خودی مشتق از بافت چربی و مقایسه آن با سلول‌های بنیادی مشتق از مغز استخوان صورت گرفت.روش‎کار: پس از ایجاد زخم‌های باز تجربی در اندام‌های حرکتی چهار اسب و درمان آن‌ها با استفاده از سلول‌های بنیادی مزانشیمی خودی، از روش Real-Time PCR برای ارزیابی و مقایسه میزان بیان ژن‌های کلاژن تیپ 1 و 3 در روند التیام زخم استفاده شد.نتایج: اختلاف معنی‌دار در تغییر بیان ژن‌های کلاژن تیپ 1 و 3 در بین گروه‌های درمانی مشاهده گردید. با وجود آن که بیشترین میزان بیان ژن‌های انواع کلاژن مربوط به سلول‌های بنیادی مشتق از مغز استخوان بود اما بین این سلول‌ها و سلول‌های مشتق از بافت چربی اختلاف معنی‌داری مشاهده نشد.نتیجه­گیری نهایی: بر این اساس با توجه به مزایای سلول‌های بنیادی مزانشیمی مشتق از بافت چربی و عملکرد قابل قبول، این سلول‌ها می‌توانند به عنوان روشی نوین در تسریع فرایند التیام زخم‌های اندام حرکتی مد نظر قرار گیرند.}, keywords_fa = {ترمیم زخم,اسب,سلول‌های بنیادی مزانشیمی,مغز استخوان,بافت چربی}, url = {https://jvr.ut.ac.ir/article_86083.html}, eprint = {https://jvr.ut.ac.ir/article_86083_586fea0a6b2785b02feb353a169e2127.pdf} }
