بررسی سرواپیدمیولوژیک هلیکوباکتر پیلوری در افراد مرتبط و غیر مرتبط با سگ و گربه در اهواز (1394-1393)
زمینه مطالعه: هلیکوباکتر پیلوری یکی از شایعترین عفونتهای میکروبی در انسان است که با زخم معده و دوازدهه، التهاب و سرطان معده ارتباط دارد. هلیکوباکتر پیلوری از از طریق آب و غذا و راه مدفوعی- دهانی و دهانی- دهانی منتقل میشود و در معده و روده انسان و چند گونه حیوانی از جمله سگ و گربه کلونیزه میشود و بنابراین احتمال انتقال این عفونت از حیوانات به انسان نیز وجود دارد. هدف: هدف از این مطالعه بررسی شیوع سرمی آلودگی به هلیکوباکتر پیلوری در افراد مرتبط و غیر مرتبط با سگ و گربه در اهواز و همچنین ارتباط آن با فاکتورهای میزبانی و محیطی بود. روش کار: نمونههای سرمی از 356 نفر، شامل 147 نفر افراد مرتبط و 209 نفر افراد غیر مرتبط با سگ و گربه جمعآوری گردید و به روش الیزا مورد ارزیابی قرار گرفت. نتایج: شیوع سرمی ظاهری و واقعی هلیکوباکتر پیلوری در افراد تحت بررسی به ترتیب 4/58% ( فاصله اطمینان 95%: 5/63-3/53 د٪) و 53/44 ٪ (فاصله اطمینان95%: 73/49-33/39 ٪) بود. شیوع ظاهری در افراد در تماس با سگ و گربه 1/72% ( فاصله اطمینان 95%: 4/79-8/64 ٪) و در افراد بدون تماس 8/48% ( فاصله اطمینان 95% 6/55-42 ٪) بود (001/0>p). شانس آلودگی در افراد در تماس با سگ و گربه 71/2 برابر افراد بدون تماس (فاصله اطمینان 95%: 26/4- 73/1 ) بود (001/0>p). این بررسی نشان داد که تماس با سگ و گربه، تحصیلات، سابقه زخم معده و اختلالات گوارشی و وضعیت تأهل ارتباط معنیداری با آلودگی دارد. نتیجهگیری نهایی: مطالعه حاضر نشان داد که تماس با سگ و گربه فاکتور خطر برای آلودگی انسان به هلیکوباکتر پیلوری میباشد و باید اقدامات کنترلی و پیشگیرانه مد نظر سیاستگذاران بهداشتی قرار گیرد.

اثر افزودن خوراکی عصاره پوست انار و آنتیاکسیدان تجاری بر عملکرد، گوارشپذیری مواد غذایی، فلور میکروبی و تیترآنتیبادی جوجههای گوشتی
زمینه مطالعه: استفاده ازآنتیاکسیدانهای تجاری در جیره جوجههای گوشتی دارای چربی به دلیل سرطانزا بودن، تورم جگر و تغییر در فعالیت آنزیمها محدود شده است. جایگزینهای گیاهی دارای خواص آنتیاکسیدانی و ضدمیکروبی،با افزایش خوشخوراکی، بهبود عملکرد دستگاه گوارش و بهبود در سیستم ایمنی سبب بهبود عملکرد پرندگان شدهاند. هدف: بررسی اثر افزودن عصاره پوست انار به جیره دارای روغن سویا بر عملکرد و تیتر آنتیبادی جوجههای گوشتی و مقایسه عصاره پوست انار با آنتیاکسیدان تجاری، هدف این آزمایش بود. روشکار: این پژوهش در قالب آزمایش فاکتوریل 2×2×2 با سه عامل عصاره پوست انار، آنتیاکسیدان تجاری و روغن سویا بر پایهی طرح کاملاًً تصادفی انجام شد. سیصد و بیست قطعه جوجه گوشتی 11 روزه سویه راس 308 به طور تصادفی به هشت تیمار (دارای چهار تکرار و هر تکرار شامل ده پرنده).اختصاص داده شدند. دادهها با رویه GLM نرم افزار SAS واکاوی، و میانگین حداقل مربعات گروهها در سطح معنیداری 5 ٪ مقایسه شدند. نتایج: روغن سویا سبب نا مطلوب شدن ضریب تبدیل خوراک در دوره رشد و کل دوره شد و فلور میکروبی مضر دستگاه گوارش را افزایش داد. عصاره پوست انار موجب بهبود افزایش وزن روزانه و خوراک مصرفی در کل دوره شد ولی بر ضریب تبدیل خوراک اثری نداشت. عصاره پوست انار سبب بهبود گوارش پذیری مواد مغذی، فلور میکروبی مفید لاکتوباسیلوس و افزایش تیتر آنتیبادی در 39 روزگی شد. آنتیاکسیدان تجاری بر افزایش وزن روزانه، خوراک مصرفی روزانه، ضریب تبدیل خوراک و تیتر آنتیبادی اثری نداشت و فلور میکروبی مفید دستگاه گوارش را کاهش داد. نتیجهگیرینهایی: به طور کلی نتایج این پژوهش نشان داد که عصاره پوست انار در جیرههای با و بدون چربی میتواند با بهبود خوراک مصرفی روزانه، گوارش پذیری مواد غذایی، فلور میکروبی مفید و سیستم ایمنی، بدون اینکه تأثیر نامطلوبی بر ضریب تبدیل خوراک مصرفی داشته باشد، سبب افزایش وزن روزانه پرندهها شود.

تأثیر سطوح مختلف تریپتوفان بر عملکرد، خصوصیات لاشه و برخی فراسنجه های خونی جوجه های گوشتی تحت تنش گرمایی
زمینه مطالعه: تنش گرمایی از طریق تغییرات فیزیولوژیک در بدن باعث کاهش عملکرد در طیور میگردد. هدف: این تحقیق به منظور بررسی تأثیر سطوح مختلف تریپتوفان بر عملکرد، خصوصیات لاشه و برخی فراسنجههای خونی جوجههای گوشتی تحت تنش صورت گرفت. روش کار: دویست و پنجاه قطعه جوجه گوشتی نر یکروزه سویه تجاری راس 308 در قالب طرح کاملاً تصادفی با 5 تیمار و 5 تکرار (پن) به ازای هر تیمار و10 جوجه در هر تکرار پرورش داده شد. جوجههای گوشتی گروه شاهد با جیرهی پایه و سایر تیمارها با جیرهی پایه همراه با سطوح مختلف 110، 120 ، 130و 140 % نیاز سویه راس تغذیه شدند. جیرههای آزمایشی در دوره پایانی (25-42 روزگی) و در شرایط تنش حرارتی (oC 1±32 به صورت دورهای از 9 صبح تا 5 بعد از ظهر) استفاده شدند. نتایج: نتایج نشان داد که افزایش وزن در دوره پایانی با افزایش سطح تریپتوفان به 110 % به طور خطی افزایش یافت اما افزایش سطح تریپتوفان جیره به 120، 130 و 140 % منجر به بدتر شدن افزایش وزن گردید (05/0>p). ضریب تبدیل خوراک در هر دو دوره پایانی و کل، با افزودن سطح تریپتوفان به 110 و 120 %، به صورت خطی کاهش پیدا کرد ولی سطوح بالاتر تریپتوفان (130 و 140 %) در هر دوره باعث افزایش ضریب تبدیل خوراک شدند (05/0>p). افزایش سطح تریپتوفان به 130 % نیاز سویه راس باعث افزایش خطی میزان هر سه آنزیم لاکتات دهیدروژناز، آلانین آمینوترانسفراز و آلکالین فسفاتاز گردید (05/0>p). مکملسازی جیره با سطوح مختلف تریپتوفان اثری بر اوره، پروتئین، کراتینین، آلبومین،گلوکز و کلسترول خون در 42 روزگی بجز تریگلیسرید نداشت (05/0<p). افزایش سطح تریپتوفان تا 140 % باعث کاهش میزان تریگلیسرید گردید. نتیجهگیری نهایی: به طور کلی، نتایج آزمایش اخیر نشان میدهد که سطوح تریپتوفان 110 و 120 % نیاز سویه راس بهترین سطوح برای بهبود عملکرد جوجههای تحت تنش گرمایی است.

اثر تغذیه با آغوز گرمادهی شده بر میزان جذب ایمونوگلوبولین G و پروتئین تام سرم در گوساله های شیری تازه متولد شده
زمینه مطالعه: یکی از روشهای کاهش یا حذف باکتریهای بیماریزای آغوز، استفاده از روش گرمادهی است. هدف: این مطالعه به منظور بررسی اثر گرمادهی آغوز گاو بر انتقال ایمنی غیر فعال در گوسالههای نوزاد انجام پذیرفت. روش کار: ابتدا با جمع آوری L 96 آغوز دوشش اول از گاوهای هولشتاین چند شکمزا، یک بانک آغوز همگن تهیه شد، سپس 24 رأس گوساله نوزاد قبل از دریافت آغوز در 4 گروه درمانی مورد مطالعه قرارگرفتند. به گروه اول آغوز خام (6رأس)، به گروه دوم آغوز گرمادهی شده در دمای ºC60 به مدت min30 (6رأس)، به گروه سوم آغوز گرمادهی شده در دمای ºC60 به مدتmin 60 (6رأس) و به گروه چهارم آغوز گرمادهی شده در دمای ºC60 به مدت min 90 (6 رأس) خورانده شد. گوسالهها در نوبت اول L 2 و در نوبت دوم نیز L 2 آغوز را به ترتیب 2 و 12 ساعت بعد از تولد به وسیله بطری پستانکدار دریافت نمودند. به منظور شمارش باکتریایی تام و تعیین غلظت IgG آغوز، قبل و بعد از گرمادهی از بانک آغوز نمونه برداری شد. جهت بررسی غلظت سرمی پروتئین تام و IgG در ساعتهای صفر (قبل از خوردن آغوز)، 6، 24، 48 و 72 (پس از خوردن آغوز) از گوسالهها خونگیری به عمل آمد. نتایج: غلظت پروتئین تام و IgG سرم در گروههای مختلف درمانی اختلاف معنیداری با یکدیگر نداشتند، اما توان ظاهری جذب IgG در ساعت 6 در گوسالههایی که آغوز گرمادهی شده دریافت نموده بودند در مقایسه با گروهی که آغوز خام دریافت نموده بودند به طور معنیداری بیشتر بود. نتیجهگیری نهایی: گرمادهی مخزنی آغوز در سطح گاوداری و در دمای ºC60 تا min 90 تأثیری بر غلظت سرمی IgG نداشت.

تأثیر الکارنیتین و ویتامین C بر شاخصهای رشد، ترکیب بدن و فراسنجه های بیوشیمیایی سرمی در بچه ماهی قزل آلای رنگین کمان
زمینه مطالعه: بیوسنتز داخلی کارنیتین مستلزم وجود اسیدهای آمینه متیونین، لیزین، ویتامینهای نیاسین و اسیداسکوربیک میباشد و استفاده از الکارنیتین و ویتامین C ممکن است بتواند اثرات الکارنیتین به تنهایی را بهبود بخشد. هدف: در این مطالعه، تأثیر سطوح مختلف الکارنیتین و ویتامین C بر شاخصهای رشد، ترکیب بدن و فراسنجههای سرمی در بچهماهی قزلآلای رنگینکمان بررسی شد. روش کار: بدین منظور، 540 قطعه بچهماهی با میانگین وزنی g 2±40، در تیمارهای آزمایشی حاوی ویتامین C در سه سطح صفر، 500، 1000 و الکارنیتین در دو سطح صفر و 800 (mg/kg جیره) با سه تکرار قرار گرفتند. نتایج: نتایج نشان داد که افزایش وزن، ضریب رشد ویژه و ضریب تبدیل غذایی در تیمار با ویتامین C 500 و الکارنیتین mg 800 نسبت به تیمار فاقد مکمل (شاهد) افزایش معنیداری (05/0>p) یافت. بیشترین و کمترین میزان چربی بهترتیب در تیمار شاهد و در تیمار حاوی ویتامینC 1000 و الکارنیتین mg/kg 800 جیره مشاهده شد (05/0>p). بیشترین مقدار پروتئین لاشه در تیمار حاوی ویتامین C 500 و الکارنیتین 800 مشاهده گردید (05/0>p). کلسترول و گلوکز نیز تحت تأثیر ترکیب ویتامین C 500 و الکارنیتین mg 800، کاهش معنیداری (05/0>p) یافت، ترکیب ویتامین C 1000 و الکارنیتین mg 800، موجب کاهش معنیداری در میزان تریگلیسرید خون گردید (05/0>p). نتیجهگیری نهایی: در نهایت مشخص شد که استفاده از ویتامین C به میزان 500 و ال-کارنیتین به میزان mg/kg 800 در جیره غذائی ماهی قزلآلای رنگینکمان میتواند موجب بهبود فراسنجههای رشد گردد.

اثر سطح مصرف و اندازه ذرات پوسته جو بر مورفولوژی و جمعیت باکتریایی روده جوجه های گوشتی
زمینه مطالعه: فیبر نامحلول موجود در خوراک طیور اغلب به عنوان عاملی نامطلوب در نظر گرفته میشود که ارزش غذایی جیره را کاهش میدهد، اما شواهدی نیز در دست است که این نوع فیبر باعث بهبود صفات تولیدی میشود. هدف: پوسته خارجی جو از نظر اثر بر عملکرد تولیدی جوجه گوشتی مورد بررسی قرار گرفت. روش کار: در این آزمایش از 200 قطعه جوجه گوشتی از سویه راس 308 در قالب یک طرح کاملاً تصادفى با 5 تیمار و 4 تکرار استفاده شد. جیرهها شامل جیره شاهد فاقد پوسته جو و چهار جیره آزمایشی حاوی پوسته جو در دو سطح 75/0% یا 5/1%، هر یک در دو اندازه کوچکتر از mm1 و یا بین mm1 تا 2 بودند. نتایج: نوع جیره اثری بر میزان مصرف خوراک در گروههای آزمایشی نداشت. بهترین افزایش وزن زنده و ضریب تبدیل خوراک در جوجههای گوشتی تغذیه شده با جیره حاوی 5/1% پوسته جو با اندازه ذرات mm2-1 ثبت شد و در کل دوره آزمایش، تفاوت مشاهده شده با گروه شاهد معنیدار بود (05/0>p). مصرف فیبر منجر به افزایش تعداد سلول گابلت در ژژنوم در مقایسه با گروه شاهد شد (05/0>p). بر خلاف انتظار، مصرف پوسته جو باعث کاهش ارتفاع پرز در مقایسه با گروه شاهد در همه بخشهای روده کوچک شد (05/0>p). در ژژنوم، عمق کریپت جوجههای شاهد بیشتر از جوجههای تغذیه شده با 5/1% پوسته جو با اندازه ذرات کوچکتر از 1 mm بود (05/0>p). نسبت عمق کریپت به ارتفاع پرز در ژژنوم جوجههای شاهد بیشتر از پرندههای تغذیه شده با جیره حاوی گروه 5/1% پوسته جو با اندازه ذرات کوچکتر از mm1 بود (05/0>p). در ایلئوم، جوجههای تغدیه شده با جیره شاهد حاوی تعداد کمتری لاکتوباسیلوس و تعداد بیشتری اشریشیاکولی در مقایسه با اکثر تیمارهای حاوی پوسته جو بودند (05/0>p). نتیجه گیری نهایی: سطح 5/1 ٪ پوسته جو با اندازه ذرات mm2-1 موجب بهبود صفات تولیدی جوجههای گوشتی گردید، ضمن آنکه وارد نمودن مقادیر تا 5/1% از پوسته جو به عنوان یک منبع لیگنوسلولزی به جیره جوجههای گوشتی باعث تحریک رشد لاکتوباسیلوسها و کاهش تعداد اشریشیاکولی در مجرای گوارش شد و این تغییر ترکیب میکروفلور روده پیشنهاد کننده اثر پری بیوتیکی پوسته جو در مجرای گوارش جوجههای گوشتی است.

مطالعه اثر نانوکپسولاسیون عصاره چای سبز بر زنده مانی لاکتوباسیلوس کازئی و بیفیدوباکتریوم لاکتیس در بستنی سیمبیوتیک
زمینه مطالعه: علیرغم فواید بسیار ترکیبات طبیعی از جمله عصاره چای سبز، استفاده از آنها در تولید مواد غذایی و دارویی کاربرد محدودی دارد. امروزه کپسولاسیون ترکیبات طبیعی در نانولیپوزومها به عنوان یک روش مناسب و کارامد جهت محافظت این مواد طی فرایند تولید و نگهداری، مورد استفاده قرار میگیرد. هدف: در این مطالعه، خصوصیات فیزیکوشیمیایی نانولیپوزومهای عصاره چای سبز و فعالیت پری بیوتیکی آن بر روی دو پروبیوتیک لاکتوباسیلوس کازئی و بیفیدوباکتریوم لاکتیسدر محیط کشت و بستنی بررسی شد. روش کار: ابتدا عصاره آبی چای سبز به روش فیلم نازک کپسوله شده و توسط امواج مافوق صوت به اندازههای نانو در آمدند. به منظور بررسی خواص پری بیوتیکی نانولیپوزومهای عصاره چای سبز، 1 % از این عصاره در حضور دو پروبیوتیک لاکتوباسیلوس کازئی و بیفیدوباکتریوم لاکتیس به محیط کشت و بستنی اضافه شدند. نتـایـج: میانگین قطر نانولیپوزومهای بدست آمده، حدود nm 9/1 ±7/44 با شاخص پراکندگی 014/0 ± 203/0، و میزان محصور سازی عصاره چای سبز نانولیپوزومی تحت شرایط بهینه 97% بود. علاوه بر این، افزودن 1% عصاره چای سبز نانولیپوزومی در محیط کشت، رشد لاکتوباسیلوس کازئی و بیفیدوباکتریوم لاکتیس را به طور معنیداری افزایش داد. در بین گروههای مورد مطالعه، کمترین کاهش در میانگین لگاریتم تعداد لاکتوباسیلوس کازئی در طول مدت زمان نگهداری در بستنیهای حاوی 1% عصاره چای سبز نانولیپوزومی معادل log 59/1 و بیشترین کاهش در بستنیهای گروه کنترل برابر با log 16/4 مشاهده شده است. همچنین کمترین اختلاف در میانگین لگاریتم تعداد بیفیدوباکتریوم لاکتیس در بستنیهای حاوی 1% عصاره چای سبز نانولیپوزومی معادل log 84/0 و بیشترین اختلاف در بستنیهای گروه کنترل برابر با log 4/4 بدست آمده است. نتیجه گیری نهایی: نانو کپسولاسیون عصاره چای سبز اثر معنیداری در رشد و زنده مانی پروبیوتیکها در محیط کشت و بستنی دارد و میتوان از این روش به منظور افزایش بقای پروبیوتیکها طی فرایند تولید و نگهداری مواد غذایی استفاده کرد.

بررسی شیوع آلودگی به انگلهای مونوژن در 100 قطعه گلدفیش مراکز توزیع شهر تهران (Carassius auratus Linnaeus, 1758)
زمینه مطالعه: گلدفیش (Carassius auratus) از کپور ماهیان Cyprinidae، یکی از پرطرفدارترین ماهیان آکواریومی در ایران و جهان است. قدمت تجارت ماهیان زینتی به ویژه گلدفیش به بیش از صد سال میرسد.اگر بر این باور باشیم که هر گونه موجود آبزی و هر کشوری فون بیماریزای خاص مشخص و نیز ناشناختهای دارا میباشد،خطرات احتمالی انتقال این عوامل بیماریزا توسط وارادات ماهیان زینتی (چه در سطح کشوری، استانی، شهری) را بیشتر آشکار میسازد. با این وجود تحقیقات محدودی در زمینه آلودگی انگلی آنها صورت گرفته است. هدف: بنابراین شناسایی و بررسی عوامل بیماری زای ماهیان به ویژه انگلها قبل از نقل و انتقال ماهیان به محیط جدید به منظور کنترل و پیشگیری از نقل و انتقال ماهیان آلوده و به تبع آن انگلها به محیط جدید از اهمیت ویژهای برخوردار است. روش کار: در این بررسی پوست و آبشش 100 عدد ماهی گلدفیش، پس از انتقال به آزمایشگاه دانشکده دامپزشکی دانشگاه تهران، از نظر ظاهری بررسی شدند. سپس با تهیه لام مرطوب از پوست و آبشش ، از نظر آلودگی به انگلهای مونوژن مورد بررسی دقیق قرار گرفتند. نتایج: در مجموع تعداد 9 گونه انگل از 3 خانواده داکتیلوژیریده، ژیروداکتیلیده و انسیلودیسکوئیده، شامل؛ 4 گونه ژیروداکتیلوس (ژیروداکتیلوس کوبایاشی، ژیروداکتیلوس گورلئی و ژیروداکتیلوس sp.)، 4 گونه داکتیلوژیروس (داکتیلوژیروس واستاتور، داکتیلوژیروس فورموزوس، داکتیلوژیروس بائری و داکتیلوژیروس sp.) و 1 گونه آنسیلودیسکوئیدس sp. از ماهیان مذکور جدا سازی شده و شناسایی گردیدند. ژیروداکتیلوس کوبایاشی در بین انگلهای یافت شده شایعترین انگل (%10) بود و بعد از آن داکتیلوژیروس واستاتور (%9) قرار داشته است. نتیجه گیری نهایی: اگرچه مقاوت این ماهیان در برابر عوامل بیماری زا بالا است، اما نقل و انتقال این ماهیان به یک بوم سازگان آبی جدید، میتواند عامل انتقال عوامل بیماریزا باشد و باعث شیوع بیماریهای جدید در محیط تازه شده و با ایجاد همه گیری در بین ماهیان سالم خسارات جبران ناپذیری را ایجاد نماید.

زمینه مطالعه: کروناویروسها طیف گستردهای از بیماریهای تنفسی، رودهای، کبدی، عصبی با حدتهای مختلف در گربه، سگ، خوک، جوندگان،گاو، پرندگان و انسان ایجاد میکنند. توانایی بالای کروناویروسها در جهش و نوترکیبی به آنها این امکان را میدهد که در میزبانهای مختلف و شرایط اقلیمی متفاوت، فعال باشند. با توجه به احتمال وجود گاماکروناویروس در جمعیت پرندگان کشور از جمله کبوتران و همچنین عدم وجود اطلاعات کافی در این مورد، انجام مطالعهای به منظور جداسازی و شناخت خصوصیات مولکولی کروناویروس کبوترها ضروری به نظر میرسید. هدف: هدف از این مطالعه ردیابی مولکولی کروناویروس پرندگان درجمعیت کبوتران استان تهران بود. روش کار: از 25 گلههای کبوتر تعداد 250 سواب نای و 250 سواب کلواک، 20 نمونه از هر گله در سال 1393 در محدوده استان تهران به صورت تصادفی به منظور ردیابی گاما کرونا ویروس جمع آوری گردید. پس از استخراج RNA، ردیابی ویروس با استفاده از پرایمرهای ژنهای نوکلئوکپسید (N) و ناحیه غیر قایل ترجمه ناحیه ‘3 (’UTR3) و با استفاده از کیت One-step RT-PCR شرکت کیاژن انجام گرفت. نتایج: نمونه نایی در یک گله کبوتر از 25 گله مورد مطالعه (4٪) مثبت گردید. نتیجه گیری نهایی: نتایج این مطالعه حضور گاما کروناویروس در جمعیت کبوتران ایران را اثبات و قسمتی از نقشه اپیدمیولوژی این ویروس را تکمیل نمود. همچنین با توجه به میزان پایین شیوع عفونت گاما کروناویروس در کبوتر ، پیشنهاد میشود نمونهگیری از کبوترهایی که دارای علایم بالینی از جمله علایم تنفسی و گوارشی هستند و همچنین از کبوترهایی که در تماس باسایر گونههای پرندگان و یا نزدیک به مرغداریها میباشند، صورت بگیرد.

بررسی مولکولی ژنهای حدت stx1 ,stx2 ,eaeA و hlyA در باکتری اشریشیا کلی جدا شده از نمونه های کلواکی در کبوتران وحشی و تعیین مقاومت آنتی بیوتیکی آنها
زمینه مطالعه: کبوترها میتوانند حامل پاتوژنهای انسانی و حیوانی باشند که یکی از مهمترین این پاتوژنها باکتری اشریشیا کلی است. این باکتری عامل ایجاد بیماریهای مختلفی در انسان میباشد. هدف: هدف این مطالعه بررسی فراوانی حضور باکتری اشریشیا کلی در ناحیه کلواک کبوتران شهر تهران و بررسی حضور ژنهای حدت و همچنین الگوی مقاومت آنتیبیوتیکی جدایهها میباشد. روش کار: در مجموع 117 نمونه مدفوع از ناحیه کلواک کبوتر اخذ گردید. شناسایی باکتری با کشت بر روی محیطهای افتراقی صورت گرفت. سپس برای جدایهها تست حساسیت آنتیبیوتیکی به روش انتشار در آگار انجام شد. حضور ژنهای حدت stx1، stx2، eaeA و hlyA در جدایهها با روش Multiplex PCR تعیین گردید. نتایج: تتراسایکلین و سولفامتوکسازول به ترتیب با 5/83% و 6/85 % مقاومت، غیرمؤثرترین آنتیبیوتیکها بودند و هیچ کدام از جدایهها به آنتیبیوتیک کوآموکسیکلاو مقاوم نبودند. فراوانی ژن stx1، stx2 و hly به ترتیب 09/3 ، 18/6 و 06/2 گزارش شد. ژن hlyA در هیچکدام از جدایهها حضور نداشت. نتیجه گیری نهایی: تناوب ژنهای stx1 و stx2 در حیوانات و پرندگان و ارتباط آنها با ایجاد بیماری در انسان، در ایران به خوبی مشخص نشده است، بنابر این ارتباط نزدیک انسان با پرندگانی مانند کبوتر و حضور سویههای مولد شیگا توکسین در این پرندگان به ظاهر سالم، نیاز به بررسی دقیق تر و جامعی برای شناسایی سویههای مولد شیگاتوکسین در جهت کنترل و پیشگیری از انتشار مستقیم و غیر مستقیم این عامل مشترک بین انسان و دام میباشد.

بررسی موقعیت گره سینوسی دهلیزی و سرخرگ تغذیه کننده آن در اسبچه خزر
زمینه مطالعه: اسبچه خزر نژادی از اسبهای کوچک جثه بوده که متعلق به ایران میباشد. انقباضات ریتمیک قلب وابسته به دستگاه هدایتی است که شامل گره سینوسی، گره دهلیزی بطنی، باند دهلیزی بطنی و رشتههای پورکینژ میباشد. هدف: با توجه به اهمیت گره سینوسی دهلیزی در تنظیم ریتم انقباضی ضربان قلب نبودن سابقه مطالعه بر روی وضعیت این گره در اسبچه خزر اهمیت انجام این مطالعه را مشخص میسازد. روش کار: در این مطالعه از قلب 6 راس اسبچه خزراستفاده گردید. به منظور بررسی موقعیت گره سینوسی دهلیزی و سرخرگ تغذیه کننده آن از تزریق ژلاتین کروم اپاک به داخل سرخرگهای کرونری و همچنین قالب گیری از این سرخرگها توسط ردوپاس کمک گرفته شد و با مشخص نمودن شیار انتهایی، محدوده گره سینوسی دهلیزی بطور ماکروسکوپی مشخص و سپس به کمک مقاطع بافت شناسی تهیه شده از این محدوده محل آن به دقت مشخص و مورد بررسی قرار گرفت. نتایج: این گره در اسبچه توده بسیار کوچکی ازماهیچه قلبی تغییر یافته است که در زیر اپیکارد دهلیز راست در محل باز شدن سیاهرگ میانخالی قدامی در محدودهای بین سیاهرگ میانخالی قدامی و گوشک راست قرار گرفته است و سرخرگ تغذیه کننده آن بصورت چند شاخه در این گره قرار گرفته است. نتیجه گیری نهایی: سرخرگ تغذیه کننده گره سینوسی دهلیزی در واقع شاخه گوشکی راست منشعب از شاخه پروگزیمال دهلیزی چپ میباشد که خود از شاخه چرخشی سرخرگ کرونری چپ منشعب میگردد که از این نظر با اسب مطابقت دارد.

بررسی اثر عصاره هیدروالکلی گیاه اسطوخدوس (Lavandula officinalis) براعمال حرکتی در موش صحرایی نر مدل شبه بیماری پارکینسون
زمینه مطالعه: بیماری پارکینسون یک اختلال ناشی از تخریب عصبی است که در حدود 1% از جمعیت بالای 50 سال اثر میگذارد. به طور کلاسیک پارکینسون به عنوان یک بیماری مربوط به سیستم حرکتی در نظر گرفته میشود و تشخیص آن بر مبنای گروهی از علائم حرکتی نظیر سفتی عضلات، کندی حرکت و اختلال در رفلکسهای حرکتی و وضعی بنا نهاده میشود. هدف: هدف از این مطالعه ارزیابی اثرات عصاره هیدروالکلی گیاه اسطوخدوس بر اختلالات حرکتی موشهای صحرایی مدل شبه بیماری پارکینسون است. روش کار: موشهای صحرایی نر نژاد ویستار به چهار گروه تقسیم شدند (7=n). علائم بیماری پارکینسون قبل از تجویز عصاره اسطوخدوس به وسیله تزریق درون صفاقی (ip) رزرپین (mg/kg 5 ) ایجاد شد. سه گروه از موشهای صحرایی مقادیر مختلف عصاره اسطوخدوس (mg/kg, po 400، 300، 200 ) و گروه شاهد نیز به مدت 6 روز و روزی دوبار به همان حجم آب مقطر دریافت کردند. در روز هفتم، سختی عضلات و دیگر علائم پارکینسون به وسیله بار تست، روتارود و میدان باز اندازه گرفته شد. نتایج: در این مطالعه بهبود علائم ناشی از بیماری پارکینسون با استفاده از عصاره هیدروالکلی اسطوخدوس مشاهده نشد و اختلالات حرکتی آنها در مقایسه با گروه شاهد کاهش نیافت. نتیجه گیری نهایی: عصاره هیدروالکلی اسطوخدوس بر اختلالات حرکتی ناشی از بیماری پارکینسون بی تأثیر است.

بررسی تأثیر غلظتهای مختلف شوری بر بافت آبشش ماهی بنی (Barbus sharpeyi)
زمینه مطالعه: ماهی بنی از خانواده ی کپورماهیان از گونههای مهم و تجاری در منطقه جنوبی ایران و خلیج فارس محسوب شده و از لحاظ تغذیهای و اقتصادی بسیار حائز اهمیت میباشد. هدف: با توجه به اهمیت سازگاری ماهیان با شوریهای مختلف در محیطهای متفاوت و قدرت تنظیم یونی و اسمزی و حفظ هموستاز بدن مطالعه اخیر صورت گرفته است. روش کار: برای انجام این پژوهش تعداد 144 قطعه ماهی بنی سالم با میانگین وزنی g 36/2 ± 350 و طول Cm 25/1± 25 در پنج گروه مورد مطالعه قرار گرفتند. گروه اول به عنوان شاهد در آب معمولی شهری کلرزدایی شده و چهار گروه بعدی به ترتیب در غلظتهایی شوری ppt 4، 8، 12و 16 در شرایط یکسان نگهداری شدند. در روزهای 1، 3، 7، 14، 21 و 28 از کمان آبششی دوم از سمت چپ نمونههایی به ضخامت حداکثر cm 5/0 تهیه و در محلول بوئن قرار داده شدند. سپس به روش استاندارد و معمول تهیه مقاطع بافتی برشهایی به ضخامت µm 6- 5 تهیه و رنگآمیزی H&E بر روی آنها انجام گرفت. نتایج: نتایج حاصل از این تحقیق نشان داد که انتقال تدریجی ماهی بنی به آب با شوری بالا باعث تغییرات واضح در تعداد و نحوه توزیع سلولهای کلراید در روزهای مختلف نمونه برداری همراه بوده است. بویژه این تغییرات در اندازه این سلولها در دو موضع فیلامنتی و لاملایی مشهود بوده است. نتیجه گیری نهایی: با توجه به روند کاهشی منابع آب شیرین از این تحقیق میتوان نتیجه گرفت که ماهی بنی تا چه اندازه میتواند بدون تغییر در ساختار آبشش در مقابل شوری مقاومت کند. این یافتهها نشان داد که ماهی بنی شوری پسند بوده و غلظت g/l 4 برای آن بهینه و غلظتهای g/l 12 و 8 برای آن قابل تحمل میباشد.

مقایسه تأثیرتجویز خوراکی و تزریقی عصاره گیاه سرخارگل (Echinacea purpurea) بر برخی شاخصهای خونی و ایمنی ماهی کپور علفخوار (Ctenopharyngodon idella)
زمینه مطالعه: محرکهای ایمنی با منشأ گیاهی در آبزی پروری دارای مزایای زیادی نسبت به واکسنها و آنتی بیوتیک درمانی هستند. هدف: در این تحقیق اثر تجویز خوراکی و تزریقی سرخارگل (Echinacea purpurea) بر برخی پارامترهای ایمنی و خونی ماهی کپور علفخوار ارزیابی شد. روش کار: بدین منظور 240 عدد ماهی آمور به چهار گروه در سه تکرار تقسیم شدند. تیمار اول عصاره اکیناسه را به میزانmg/kg.b.w 400 به روش i.p دریافت نمودند. تیمار دوم میزان 5/0% پودر سرخارگل در خوراک روزانه خود دریافت نمودند. تیمار سوم و چهارم نیز بترتیب، به عنوان کنترل تزریقی و کنترل خوراکی در نظر گرفته شدند. ماهیها به مدت 40 روز نگهداری شده و در روزهای صفر، 10، 20، 30 و 40 از ماهیها خونگیری شده و شاخصهای ایمنی سرم شامل: میزان فعالیت لایزوزیم، قدرت باکتریکشی، فعالیت کمپلمان و احیای نیتروبلوتترازولیوم (NBT) و شاخصهای خونی شامل: هماتوکریت، هموگلوبین، تعداد گلبولهای قرمز و تعداد و نسبت گلبولهای سفید بررسی گردیدند. در روز 40 تیمارها با باکتری آئروموناس هیدروفیلا مواجهه داده شدند. نتایج: نتایج نشان داد که میزان فعالیت لایزوزیم سرم، قدرت باکتری کشی سرم، فعالیت کمپلمان و احیای NBT در تیمار تزریقی افزایش معنیداری نسبت به تیمار شاهد داشت (0.05>p) درحالیکه در تیمار خوراکی افزایش قدرت باکتری کشی سرم و احیای NBT در روز 30 و 40 مشاهده گردید (0.05>p). پارامترهای خونی مرتبط به گلبولهای قرمز در هیچکدام از تیمارها تفاوت معنیداری را نشان ندادند (0.05<p)، لیکن تعداد لوکوسیتها و نسبت هتروفیلها در تیمار تزریقی افزایشی را نسبت به تیمار شاهد نشان دادند (0.05>p). تلفات بعد از چالش در تیمار تزریقی کاهش معنیداری نسبت به تیمار کنترل داشت. نتیجهگیرینهایی: عصاره سرخارگل به عنوان کاندیدی برای تحریک ایمنی غیر اختصاصی ماهی مناسب به نظر میرسد.

تعیین آنکسین1،۲ A در سرم گوسالههای مبتلا به پنومونی تجربی با باکتری پاستورلا مولتیسیدا
زمینه مطالعه: آنکسینها پروتئینهای مهمی هستند که نقشهایی همانند هدایت سیگنالهای داخل سلولی، اتصال اسکلت سلولی غشاء، تکثیر، تمایز و خصوصاً عملکرد ممانعتی در فرآیند التهاب در بدن موجودات زنده را برعهده دارند. پاستورلا مولتیسیدا به عنوان معمول ترین بیماریزای باکتریایی مطرح بوده و شیوع بالایی را در پنومونیها به خود اختصاص میدهد. هدف: تعیین آنکسین آ-1 و آنکسین آ-2 در سرم گوسالههای مبتلا به پنومونی به صورت تجربی با باکتری پاستورلا مولتیسیدا. روش کار: در تحقیق حاضر 10 راس گوساله نر را در محدوده سنی دو تا چهار ماهه انتخاب و به دو گروه مساوی تقسیم نموده، به یک گروه به عنوان گروه تیمار باکتری پاستورلا مولتیسیدا به میزان ml 300 در رقت CFU/ml 109 × 2 و به گروه شاهد ml 300 نرمال سالین از راه دهان و توسط کاتتر مخصوص لاواژ در ناحیه نای تلقیح گردید. بر اساس جداول مخصوص، امتیازات بالینی ثبت گردید و حدود 18 تا 24 ساعت پس از تلقیح باکتری در گوسالههای گروه تیمار و همزمان با بروز تغییرات بالینی در این گروه، شستشوی برونشی حبابچهای جهت آزمایشات باکتری شناسی و سلول شناسی از مایعات حاصل از شستشو انجام و خون گیری از ورید وداج گوسالههای هر دو گروه انجام شده و سرم آنها با استفاده از کیت الایزای آنکسین مورد آزمایش قرار گرفتند. نتایج: میزان آنکسین آ-1 و آنکسین آ-2 در سرم خون گروه تیمار، افزایش معنیداری را (با استفاده از آزمون تی مستقل) نسبت به گروه کنترل نشان داد (05/0>p). نتیجه گیری نهایی: بنابراین به نظر میرسد آنکسین آ-1 و آنکسین آ-2 میتوانند به عنوان یک بیومارکر مهم در سرم خون برای تشخیص فرآیندهای التهابی همچون پنومونی استفاده شود.
