بررسی اثر سطوح مختلف پروبیوتیک لاکتوباسیلوس کازئی (Lactobacillus casei) بر عملکرد رشد و فعالیت آنزیمهای گوارشی ماهی شیربت (Barbus grypus)

چکیده

زمینه مطالعه: باکتری‌های اسید لاکتیک، متداول‌ترین نوع باکتری‌هایی هستند که به عنوان پروبیوتیک در آبزی‌پروری استفاده می‌شوند. هدف: در این مطالعه اثرات سطوح مختلف باکتری پروبیوتیکی لاکتوباسیلوس کازئی بر عملکرد رشد و فعالیت آنزیم‌های گوارشی بچه ماهیان شیربت ارزیابی شد. روش کار: تعداد 480 قطعه بچه ماهی شیربت  با وزن حدود 40 گرم به چهار تیمار (در سه تکرار) به صورت تصادفی تقسیم گردیدند. تیمار A، B و C به ترتیب  با جیره غذایی حاوی، ۱-CFU g  106×5 ، ۱-CFU g  107×5 ،۱-CFU g  108×5 به مدت 60 روز غذادهی شدند. گروه شاهد با جیره فاقد باکتری تغذیه گردید. پس از اتمام دوره، تیمارها به مدت 15 روز با غذای فاقد پروبیوتیک غذادهی گردیدند. شاخص‌های رشد و آنزیم‌های گوارشی در روزهای صفر، 30، 60، 75 مورد بررسی قرار گرفتند. نتایج: ضریب رشد ویژه، میزان رشد روزانه و میزان رشد نسبی، در غلظت B پس از 30 روز از شروع آزمایش، بهبود قابل توجهی پیدا کردند که با گروه کنترل اختلاف معنی‌داری داشت (0.05). فعالیت آنزیم کیموترپسین در تیمار B پس از 30 روز و همچنین فعالیت آنزیم تریپسین در تیمار C پس از 30 و 60 روز، افزایش معنی‌داری را نسبت به گروه شاهد نشان داد (0.05>p). در بقیه مراحل نمونه‌گیری و سایر آنزیم‌ها تفاوت معنی‌داری مشاهده نگردید (0.05>p). نتیجهگیرینهایی: نتایج نشان داد که میزان 107× 5 کلنی در گرم پروبیوتیک لاکتوباسیلوس کازئی در هر گرم خوراک برای دوره 30 روزه و میزان 108×5 کلنی در گرم آن برای دوره 60 روزه پرورش بهترین غلظت  محسوب می‌شوند.

BACKGROUND: Lactic acid bacteria are the most common class of bacteria used in aquaculture as probiotic. ObjectiveS: In this study the effects of various levels of Lactobacillus casei on the growth performance and digestive enzymes activity of juvenile Shirbot were evaluated. Methods: Four hundred-eighty juvenile Shirbot weighing 40 g were divided randomly in four treatments (in triplicate). Treatments of A, B and C were fed with 5×106 CFU g-1, 5×107 CFU g-1, 5×108 CFU g-1, respectively for 60 days. Control group was fed with free-probiotic diet. After the period, treatments were fed with free-probiotic diet for 15 days. Growth indices and digestive enzymes were examined on days 0, 30, 60 and 75. Results: In the Treatment B, Specific Growth  Ratio, Daily Weight Growth and Relative Growth Rate, after 30 days from the beginning of experiment improved considerably which, compared to control group had significant difference (p<0.05). Activity of chymotrypsin enzyme in treatment of group B after 30 days and similarly, trypsine in treatment of group C after 30 and 60 days, were increased significantly compared to control group (p<0.05). In the other points of sampling and enzymes, considerable difference was not seen (p>0.05). Conclusions: Results showed that 5×107 CFUg-1 of Lactobacillus casei for 30 days and 5×108 CFUg-1 for 60 days, are the best doses of probiotic.
