تعیین غلظت نیمه کشنده (LC50 ) و بررسی اثر بیهوش کنندگی اسانس اسطوخودوس (Lavandula angustifolia) در بچهماهی کپور معمولی (Cyprinus carpio)

زمینه مطالعه: در دهه اخیر تمایل به استفاده از گیاهان دارویی در صنعت آبزی‌پروری رو به افزایش نهاده است. هدف: این مطالعه، به‌منظور تعیین غلظت نیمه‌کشندگی (LC50) اسانس اسطوخودوس (Lavandula angustifolia) در طی مدت زمان h 96 و بررسی اثر بی‌هوشی آن در بچه‌ماهی کپور معمولی (Cyprinus carpio) انجام شد. روش کار: تعیین غلظت نیمه‌کشنده اسانس بر اساس استاندارد OECD و بصورت ساکن انجام شد. برای تعیین LC50، 150 قطعه بچه‌ماهی کپور معمولی با میانگین وزنی g 98/1± 96/26 در 6 گروه تیماربندی شدند. بعد از سپری‌شدن دوره سازش‌پذیری (دو هفته)، بچه‌ماهی‌ها در معرض غلظت‌های مختلف 12/71، 11/72، 11/73، 13/74 و mg/l 16/75 که به روش لگاریتمی محاسبه شدند قرار گرفتند و تغییرات رفتاری و تلفات در زمان‌های 24، 48، 72 و h 96 پس از در معرض قرار گرفتن با اسانس اسطوخودوس ثبت شد. برای تعیین اثر بی‌هوشی، بچه‌ماهیان کپور معمولی در 5 گروه 10 تایی در معرض غلظت‌های صعودی (160، 165، 170، 175 و mg/l 180) اسانس قرار گرفتند و یک گروه بعنوان شاهد در نظر گرفته شد. نتایج: غلظت نیمه‌کشندگی (LC50 ) h 96 اسانس اسطوخودوس mg/l 99/70 تعیین گردید. کوتاه‌ترین زمان برای بی‌هوشی کامل (مرحله 3 بی‌هوشی) در غلظت mg/l 170 با زمان بی‌هوشی min 35/0±74/3 و زمان احیا کامل min 55/0±91/3 بدست آمد که بهترین غلظت بی‌هوشی اسانس اسطوخودس در این آزمایش تعیین شد. نتیجهگیری نهایی: بر اساس نتایج این تحقیق اسانس اسطوخودوس می‌تواند بعنوان بی هوش‌کننده بکار برده شود.

BACKGROUND: In recent years, medicinal plants have become
an important option for use in aquaculture industry. Objectives: This study was conducted to determine mean
lethal concentration (LC50) and investigated the anesthetic effect
of Topped lavender essential oil (Lavandula angustifolia) on common carp
(Cyprinus carpio) juveniles. Methods:
Mean lethal concentration of essential oil was calculated based on the OECD
(Organization Economic Cooperation and Development) standard in static system.
For determining  LC50, 150
common carp juveniles with mean body weight of 26.96 ± 1.98 g were used in 6
groups and one control group and each group with three replications (7
juveniles were used for each replication). After Compatibility period (about 2
weeks), juveniles were exposed to different concentrations including 71.12,
72.11, 73.11, 74.13, 75.16 mg/l that were calculated by logarithmic formula.
Juvenile behavioral changes and mortality were recorded in the time period at
24, 48, 72 and 96 hours after exposed to Topped lavender essential oil.
Determination of Topped lavender essential oil anesthetic effect was done in 6
groups (with 3 replicates), as 10 fish exposed to increasing concentrations
including 0 (control), 160, 165, 170, 175 and 180 mg/l. Results: Mean lethal concentration
(LC50) after 96 hours was obtained 99.70 mg/l. The lowest time for completion
of anesthesia (stage 3 anesthesia) was obtained at a concentration of 170 mg/l
with 3.74± 0.35 min and the full recovery of this concentration was obtained at
3.91±0.55 min. The results showed that the best anaesthetic concentration of
Topped lavender essential oil was 170 mg/l. Conclusions: It seems that Topped lavender essential oil
could be used as anesthetic agent
